کلاژن چیست؟

کلاژن یکی از مهمترین پروتئین های ساختاری پوست انسان است که به پوست خاصیت نرمی و قابلیت ارتجاع می بخشد. در پوست انسان پروتئین های کلاژن مرتباً توسط فیبروبلاست ها سنتزشده و کلاژن های قدیمی و اضافی توسط آنزیم هایی به نام «ماتریس متالوپروتئیناز» (MMP) تجزیه و تخریب می شوند.
برای جلوگیری از فعالیت بیش از حد این آنزیم و نتیجتاً جلوگیری از تجزیه بیش از حد کلاژن ها، در درون یک پوست سالم این آنزیم ها در کنار مهارکننده هایشان حضور دارند. در واقع بین آنزیم های از بین برنده کلاژن و مهارکننده های طبیعی آنها یک تعادل آنزیمی برقرار است. این تعادل آنزیمی به پوست کمک می کند تا ظاهر سالم خود را حفظ نماید.
عواملی مانند سالخوردگی ، داشتن یک زندگی پراسترس یا مواجهه بیش از حد در برابر عوامل محرک محیط موجب می شوند که فعالیت این آنزیم ها اضافه شده و سلامت ماتریس برون سلولی تحت تاثیر قرار می گیرد. برقراری یک تعادل آنزیمی برای حفظ سلامت ماتریس برون سلولی بسیار مهم است.

کلاژن هفت

کلاژن نوع هفت، تشکیل دهنده فیبرهای اتصال دهنده سیستم اتصال درم به اپیدرم است. این سیستم مسئول محکم نگه داشتن اپیدرم بر روی درم است. نشان داده شده است که در هنگام سالخوردگی، تعداد این فیبرهای اتصال دهنده کاهش می یابد.

کلاژن یک

سالخوردگی و مواجهه طولانی مدت در برابر پرتوهای فرابنفش تاثیر منفی بر سنتز کلاژن نوع اول می گذارند. علاوه بر این، افزایش فعالیت آنزیم MMP-2 موجب تخریب بیشتر رشته های کلاژن نوع اول می شود. این اثر در کنار تخریب دیگر سوبستراهای آنزیم MMP-2، به از بین رفتن تدریجی سلامت مواد ماتریس برون سلولی خواهد انجامید.

تعادل سه جزئی کلاژن

همانطور که در دیاگرام شکل زیر مشاهده می شود، در پوست سالم یک تعادل سه جزئی بین رشته های کلاژن وجود دارد:

کلاژن اصلی ترین بخش ناحیه درم را تشکیل می دهد. حدود ۷۷% از وزن خشک بدون چربی درم، از کلاژن تشکیل یافته است. کلاژن بافت پوستی را حمایت می کند و مسئول سفتی پوست می باشد. کاهش سنتز این ماده موجب نازک شدن پوست و ایجاد خطوط و چروک های ریز می شود. به همین علت، نوسازی و افزایش سنتز رشته های کلاژن یکی از اصلی ترین اهداف به کارگیری فرآورده های ضدچروک و ضد پیری پوست می باشد، زیرا کلاژن نقشی اساسی در حفظ ساختار درم ایفا می نماید.

کلاژن بدن انسان، حاوی ۱۸ نوع اسید آمینه است که ۱۱ نوع از آن ها در درم دیده می شود. این پلیمر به شکل یک شبکه نامنظم از فیبرهای موج دار به هم تنیده است که به موازات سطح پوست در درم قرار گرفته اند.
پرولین یکی از آمینواسیدهای تشکیل دهنده پروتئین کلاژن است. پرولین را می توان با مواد رادیواکتیو علامت گذاری نموده و به کشت حاوی فیبروبلاست انسان افزود. به این ترتیب کلاژن های جدید سنتز شده را می توان از طریق شناسایی این ملکولهای علامت گذاری شده در درون ساختار پروتئین کلاژن مشخص نمود.
مشخصه کلاژن حضور دو اسید آمینه خاص به نامهای هیدروکسی لایسین و هیدروکسی پرولین است. با اندازه گیری میزان هیدروکسی پرولین می توان مقدار کلاژن سنتز شده توسط سلول را مشخص نمود. همچنین می توان مقدار کلاژن تخریب شده در اثر یک بیماری یا در خلال برخی فرایندهای فیزیولوژیکی را مشخص نمود.

آیا کلاژن را می توان به عنوان یکی از اجزاء محصولات آرایشی بهداشتی استفاده نمود؟

PALMITOYL PENTAPEPTIDE 3: Clinically proven to regenerate the skin's upper layers by stimulating collagen production, thus thickening the epidermis. Palmitoyl Pentapeptide-3 is a synthetic protein that is a fragment of the C-terminal portion of collagen I (the most common type of the nineteen forms of collagen) combined with palmitic acid to make it more lipophillic, to improve its stability and to enhance its affinity towards human skin. One could look at Palmitoyl Pentapeptide-3 as a man-made precursor to collagen I.
